OPTIMIZATION OF PLANT GROWTH REGULATOR’S CONCENTRATION NAA AND BAP ON THE GROWTH OF CROCODILE TONGUE EXPLANT

ABSTRACT

The use of optimal concentration of plant growth regulator, NAA and BAP on the growth of crocodile tongue, has been done in tissue culture laboratory, Faculty of Agriculture, Sarjanawiyata Tamansiswa University. The experiment is factorial 4 x 3, arranged in a completely randomized design with three replications. The first factor was concentration of NAA consisting of four levels: 0.50 ppm (N 1), 1.00 ppm (N2), 1.50 ppm (N3) and 2.00 ppm (N4). The second factor was concentration of BAP consisting of four levels : 0.50 ppm (B1), 1.00 ppm (B2), 1.500 ppm (N3) and 2.00 ppm (N4). The variables observed were : date of shoot emerge, number and height of shoots, number of leaves and roots, root length, shoot and root fresh weight, shoot – root fresh weight ratio and viability potensial of the explant.
There were interaction effect between NAA and BAP concentration on the number of roots and root fresh weight, but no interaction effects on other variables observed. The combination NAA of 0.500 ppm with BAP of 1.00 ppm gave the height number and fresh weight of roots. BAP concentration of 1.00 ppm could increase the growth of the explant.
